ABSTRACT

Objective: To investigate the detection types and aggregation of high-risk population of cardiovascular disease (CVD) in Jiangsu province and the related influencing factors to provide reference for the prevention and control of cardiovascular disease. Methods: A total of 120 211 participants were included in the investigation. Information was collected by questionnaire based survey, physical examination and biochemical tests. χ2 test and multivariate logistic regression were used for statistical analysis. Results: The detection rate of CVD high risk was 25.03%. The detection rates were 19.01%, 4.85%, 3.18% and 5.31% for hypertension, dyslipidemia, cardiovascular history and WHO assessed risk ≥20% types, respectively. Male, rural, old age, low education level, low family income, drinking, waist circumference exceeding standard, overweight and obesity were risk factors of CVD (all P<0.01). The composition ratios of aggregation of 1, 2 and ≥3 high risk types of CVD were 74.01%, 22.91% and 3.08%, respectively. With the increase of aggregation types, the correlation strength increased with age, rural residents, education level and annual family income. Conclusion: Targeted measures should be carried out according to different influencing factors for the prevention and control of CVD in Jiangsu province in order to achieve the maximum prevention and control effect with the minimum cost.

ABSTRACT

Objective@#The relationship between serum uric acid (SUA) levels and glycemic indices, including plasma glucose (FPG), 2-hour postload glucose (2h-PG), and glycated hemoglobin (HbA1c), remains inconclusive. We aimed to explore the associations between glycemic indices and SUA levels in the general Chinese population.@*Methods@#The current study was a cross-sectional analysis using the first follow-up survey data from The China Cardiometabolic Disease and Cancer Cohort Study. A total of 105,922 community-dwelling adults aged ≥ 40 years underwent the oral glucose tolerance test and uric acid assessment. The nonlinear relationships between glycemic indices and SUA levels were explored using generalized additive models.@*Results@#A total of 30,941 men and 62,361 women were eligible for the current analysis. Generalized additive models verified the inverted U-shaped association between glycemic indices and SUA levels, but with different inflection points in men and women. The thresholds for FPG, 2h-PG, and HbA1c for men and women were 6.5/8.0 mmol/L, 11.0/14.0 mmol/L, and 6.1/6.5, respectively (SUA levels increased with increasing glycemic indices before the inflection points and then eventually decreased with further increases in the glycemic indices).@*Conclusion@#An inverted U-shaped association was observed between major glycemic indices and uric acid levels in both sexes, while the inflection points were reached earlier in men than in women.

ABSTRACT

Objective:To explore the curative effect and mechanism of modified Jiegeng Yuansentang combined with conventional therapy for postoperative mucosal edema in patients with cchronic rhinosinusitis with nasal polyps due to pulmonary Qi stagnation, and to explore its mechanism of action. Method:The 90 patients were randomly divided into control group and observation group, 45 cases in each group.All patients were given conventional therapy after nasal endoscopy.Patients in control group were also given mometasone furoate combined with clarithromycin, and patients in observation group were given modified Jiegeng Yuansentang for internal administration and local irrigation.The course of the treatment was 6 weeks.The sino nasal outcome test-20 (SNOT-20), nasal situation Lund-Kennedy, rhinoconjunctivitis quality of life questionnaire (RQLQ), traditional Chinese medicine (TCM) syndrome, serum and nasal secretions inflammatory cytokines [tumor necrosis factor-alpha (TNF-<italic>α</italic>), interleukin-1<italic>β</italic> (IL-1<italic>β</italic>), interleukin-5 (IL-5),interleukin-17 (IL-17)], serum protein [aquaporin-1 (AQP-1), aquaporin-3 (AQP-3), aquaporin-5 (AQP-5), fibronectin (Fn)] were observed before and after treatment in two groups.The clinical symptoms, safety and recurrence after a follow-up of 12 months were compared between two groups. Result:The total effective rate was 97.73% (43/44) in observation group, which was higher than 80.95% (34/42) in control group (<italic>χ</italic><sup>2</sup>=4.726, <italic>P</italic><0.05).The patients were followed up for at least 12 months, and the recurrence rate was 4.65% (2/43) in observation group, which was lower than 32.35% (11/34) in control group (<italic>χ</italic><sup>2</sup>=4.129, <italic>P</italic><0.05).Compared with control group after treatment, the SNOT-20, Lund-Kennedy, RQLQ, TCM syndrome, TNF-<italic>α</italic>, IL-1<italic>β</italic>, IL-5, IL-17, AQP-5 were significantly decreased in observation group (<italic>P</italic><0.05), while AQP-1, AQP-3, Fn were significantly increased in observation group (<italic>P</italic><0.05).The incidence of adverse reactions was 2.27% (1/44) in observation group, lower than 57.14% (24/42) in control group (<italic>χ</italic><sup>2</sup>=5.243, <italic>P</italic><0.05). Conclusion:Modified Jiegeng Yuansentang can significantly improve postoperative mucosal edema in patients with chronic rhinosinusitis with nasal polyps due to pulmonary Qi stagnation, and the mechanism may be related to the regulation of inflammatory factors in serum and nasal secretions.

ABSTRACT

Objective: To explore the effect of Longdan Xiegantang on serum inflammatory factors, related proteins and immune function in patients of secretory otitis media (SOM) with liver and gallbladder wetness-heat Syndrome. Method:Totally 76 cases of SOM with liver and gallbladder wetness-heat syndrome admitted to our hospital from July 2017 to May 2018 were randomly divided into two groups, with 38 cases in each group. Control group was treated with triamcinolone acetonide and ambroxol. In addition to the therapy of control group, observation group was also treated with Longdan Xiegantang. Immunoglobulin (Ig) A, IgG, IgM, CD3+, CD4+, CD8+and NK, interleukin-1 beta (IL-1β), IL-5, IL-8, tumor necrosis factor-α (TNF-α), platelet activating factor (PAF), calcitonin (PCT) and water channel protein-1 (AQP-1), AQP-4, fiber link protein (Fn) and soluble interleukin-2 receptor (SIL-2R) levels of two groups were observed before and after treatment. Curative effect and adverse reaction were observed. Result:①Curative effect, after treatment, the total effective rate of observation group was 92.11%, which was higher than 76.32% of control group, with statistically significant differences (Z=2.108, Pα, PAF, PCT, IL-1β and IL-8 in observation group were lower than those in control group after treatment (PPP+, IgA, IgG and IgM of observation group were lower than those of control group (P+, CD4+, CD4+/CD8+ and NK were higher than those of control group (PConclusion:Longdan Xiegantang has a remarkable effect in treating patients of secretory otitis media with liver and gallbladder wetness-heat syndrome, and can restore symptoms, inhibit inflammatory response, activate cell and humeral immune system, reduce the secretion of AQP-1, SIL-2R and other proteins, and increase the secretion of AQP-4 and Fn proteins.

ABSTRACT

Due to the diversity and complexity, the change of chemical components in medicinal plant according to the time, cultivated varieties or ecological condition is difficult to recognize using traditional phytochemistry method. In order to analyze the pharmacodynamics material basis in Uighur medicinal plant Artemisia rupestris L. in an effective and comprehensive way, a plant metabolomics approach was established based on liquid chromatography-tandem mass spectrometry (LC-MS/MS). This study firstly focused on the effect of extraction solvents,redissolve solvents and ultrasonic time on the untargeted metabolomics, then the optimal preparation condition was selected according to metabolites coverage. After methodology validation, the approach was applied to acquire metabolic information in root, stem, branchlet, leaf and flower of Artemisia rupestris L. The results showed that the metabolome in flower was obviously different with the other organs. Coupling with multivariate statistical analysis, a batch of differential metabolites were picked out, in which 61 flavonoids, 97 rupestonic acid derivatives, 7 chlorogenic acids and 15 other compounds were primarily recognized according to the characteristic fragmentation rules of specific structure type and database retrieval. Additionally,the distribution characteristics of the above 180 differential metabolites was illustrated by cluster heat map. In conclusion,this study provided important information about the rational utilization of effective parts from Artemisia rupestris L.,and offered a novel strategy for quality control,variety improvement and reasonable development of medicinal plants.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of arsenic trioxide (AsO) on K562 cell proliferation by regulating cell cycle protein D1 and cyclin-dependent kinase inhibitor p27kip1.</p><p><b>METHODS</b>MTT was used to detect the effect of AsOon K562 cell proliferation, so as to screen out the appropriate drug concentration. Furthermore, the K562 cell apoptosis was observed by microscopy. The expression of CyclinD1 and p27kip1 in K562 cells treated with AsOwas analyzed by reverse transcription-polymerase chain reaction(RT-PCR), immunohistochemistry and Western blot.</p><p><b>RESULTS</b>AsOcould inhibit the proliferation of K562 cells in a dose- and time- dependent manner (r= 0.967). And the apoptosis cell number in AsOgroup was significantly higher than that in the control group(P<0.05). AsOcould markedly inhibit the expression of CyclinD1 in K562 cells(P<0.05), but the expression of P27kip1 was not significantly changed after AsOtreatment.</p><p><b>CONCLUSIONS</b>AsOcan induce K562 cell apoptosis and inhibit K562 cell proliferation by regulating the expression of CyclinD1.</p>

ABSTRACT

In this study, an analytical method was developed and used to quantify simultaneously protocatechuic acid, neochlorogenic acid, cryptochlorogenic acid and 1, 3-dicaffeoylquinic acid--four bioactive compounds contained in Fructus Xanthii using UPLC. The contents of four phenolic components of 28 batches of samples collected from different product areas and markets were determined and compared by means of this established method. The mobile phase was composed of methanol and water containing 0.1% phosphoric acid. Chromatography was monitored at dual-wavelengths--220 and 327 nm. Flow rate was 0.4 mL x min(-1) and column temperature was 35 degrees C. The correlation coefficient between concentration and chromatographic peak area of protocatechuic acid, neochlorogenic acid, cryptochlorogenic acid and 1, 3-dicaffeoylquinic acid was over 0.9999 in the range of 0.3570-35.70, 2.500-250.0, 1.060-106.1, 1.010-101.0 microg x mL(-1), respectively. The average recoveries of the four compounds were 97.68%, 99.55%, 97.92% and 100.4%, respectively. In conclusion, the established method can rapidly attain an accurate and reproducible result used to control the quality of Fructus Xanthii.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of high glucose on expression of matrix metalloproteinase-2 (MMP-2) and extracellular matrix metalloproteinase inducer (EMMPRIN) in human umbilical vein endothelial cells (HUVECs).</p><p><b>METHODS</b>HUVECs were cultured in DMEM media containing high glucose with or without polyphenols for 24 hours respectively. The activity of MMP-2 in the supernatant was detected by zymography. The expression of MMP-2 mRNA and protein in HUVECs were detected by RT-PCR and Western blot respectively. The expression of EMMPRIN mRNA and protein in the cells were determined by RT-PCR as well as immunocytochemistry and Western blot respectively.</p><p><b>RESULTS</b>The expression of MMP-2 and EMMPRIN mRNA were suppressed by a high concentration of glucose. Both the MMP-2 activity and protein level of EMMPRIN expression were also significantly decreased. Polyphenols abolished all the above changes of HUVECs induced by a high glucose level (P < 0.05).</p><p><b>CONCLUSIONS</b>An acute high glucose stimulation down-regulates the activity of MMP-2, the expressions of MMP-2 and EMMPRIN at RNA and protein levels in the endothelial cells, which may play an important roles in diabetic vascular complications in the early phase. Polyphenols treatment can diminish the detrimental effects of high glucose on HUVECs.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the impact of intracellular interaction between fibroblasts and colorectal cancer cells on the expression of insulin growth factor binding protein7 (IGFBP7).</p><p><b>METHODS</b>Colorectal cancer cells SW480 were cultured with or without fibroblasts HELF cells in RPMI 1640 medium for 0 h, 48 h, 7 2 h and 96 h, respectively. By RT-PCR and immunohistochemical staining methods,the expression of IGFBP7 was detected in mono-and co-cultured cells.</p><p><b>RESULT</b>When SW480 cells were co-cultured with HELF cells, IGFBP7 expression in SW480 cells was significantly upregulated. Furthermore, IGFBP7 was induced in HELF cells both at mRNA and protein levels, which did not express when cells were mono-cultured.</p><p><b>CONCLUSION</b>Fibroblasts-colorectal cancer cells interaction induces the expression of IGFBP7, which indicates tumor-stroma interactions may play an important role in colorectal cancer development.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of calorie restriction on the high fat diet rats mRNA expressions of liver forkhead box O1(FoxO1), phosphoenolpyruvate carboxykinase (PEPCK), glucose-6-phosphatase (G-6-P) and to explore the possible mechanisms.</p><p><b>METHODS</b>24 normal 6-week-old male Wistar rats were randomly divided into three groups: normal chow group (NC, n = 7), high fat diet group (HF, n = 9) and calorie restriction group (CR, n = 8). They were fed for 12 weeks. At the end of the experiment, the rats were sacrificed and their fasting blood glucose (FBG), insulin (INS), triglycerides (TG), total cholesterol (TC) were measured. Their visceral fat (VF) and body weight (BW) were also measured and VF/BW was calculated. Gene expression was investigated by using semi-quantitative RT-PCR methods. Liver histology was studied with HE stained slides.</p><p><b>RESULTS</b>Compared with the NC group, HF group rats developed visceral obesity which was accompanied by higher FBG, plasma INS, TG, and TC. The levels of FoxO1, PEPCK, and G-6-P increased by 18.9%, 33.8%, and 24.6%, respectively (P less than 0.01). Liver steatosis was observed with microscopy. The BW, VF FBG, INS, TG and TC of the CR group rats were lower in comparison to those of the HF group. The levels of FoxO1, PEPCK and G-6-P were lower by 26.6%, 35.0%, 34.3% (P less than 0.01). Meanwhile, liver steatosis was also milder.</p><p><b>CONCLUSION</b>Calorie restriction can inhibit the expressions of FoxO1, PEPCK and G-6-P, strengthen insulin signal conduction, suppress gluconeogenesis and thus regulate glycometabolism.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of resveratrol on the nonalcoholic fatty liver (NAFL).</p><p><b>METHODS</b>Thirty Wistar rats were allocated into three groups (10 for each): a normal control group (NC, fed with standard chow), a high fat feeding group (HF, fed with a high fat diet) and a resveratrol treated group (RT). The RT rats were fed a high fat diet all the time (as the HF) and received resveratrol administration (100 mg . kg-1. d-1) started on the 6th week of the experiment. The experiment lasted for 16 weeks. Then all the rats were sacrificed and hyperinsulinemic-euglycemic clamp was performed to evaluate insulin sensitivity. Liver histology was studied by HE stained microscopic preparations of the livers. Phosphorylation of AMP-activated protein kinase (AMPK) levels were determined by Western blot.</p><p><b>RESULTS</b>Compared to the NC, visceral fat index and liver mass index was lower and fasting serum insulin (FINS) was higher and glucose infusion rate (GIR) was lower in the HF (t=10.554, P less than 0.01). Severe liver steatosis was observed microscopically in the HF. In addition, phosphorylation of AMPK levels in the HF was 45.8% lower than that of the NC (t=8.384, P less than 0.01). Compared with the HF, the visceral fat index, liver mass index, FINS and GIR were all much lower, and the liver steatosis was milder than those in the RT. Further phosphorylation of AMPK seen was 70.2% higher in RT compared with that in the NC (t=3.816, P less than 0.01).</p><p><b>CONCLUSION</b>Resveratrol administration improved high-fat feeding induced insulin resistance and fatty liver by activating AMPK.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the prevalence of peripheral arterial disease (PAD) in China type 2 diabetic patients and to demonstrate the relationships between putative risk factors and PAD.</p><p><b>METHODS</b>In total 1,397 type 2 diabetic patients aged 50 years and older were enrolled and determined ankle-brachial index (ABI) and brachial-ankle pulse wave velocity (baPWV) in 15 Class III Grade A hospitals in 7 major cities of China.</p><p><b>RESULTS</b>Mean patient age was 63.7 +/- 8.2 years and mean duration of diabetes mellitus was 9.39 +/- 7.4 years. Two hundreds and seventy-two (19.47%) patients were diagnosed as PAD by ABI < 0.9, 122 (18.37%) in male and 150 (20.46%) in female. PAD patients had a significantly longer duration of diabetes mellitus, higher hemoglobin A1c, and a significantly lower mean body mass index than non-PAD ones. Aging, smoking, and systolic blood pressure were found to be positively related with the prevalence of PAD. In terms of lipid profiles, no variable was found to relate with PAD. Notably, baPWV showed as the same significant guiding index for PAD, almost matched with ABI.</p><p><b>CONCLUSIONS</b>PAD is a common complication in China type 2 diabetic patients. Therefore, PAD screening and treatment should be emphasized for diabetic patients with high risk factors.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expression of extracellular matrix metalloproteinase inducer gene (EMMPRIN) regulated by the interactions between fibroblasts and colon cancer cells, and to study its role in the invasion and metastasis of colon cancer.</p><p><b>METHODS</b>Colon cancer cells (SW480) were co-cultured with fibroblasts (HELF) in RPMI 1640 media for 0, 12, 24 and 48 hours. The expression of EMMPRIN in SW480 cells and HELF cells was documented by RT-PCR and immunocytochemistry.</p><p><b>RESULTS</b>The mRNA and protein expressions of EMMPRIN in SW480 cells were remarkably increased by the co-culturing with HELF cells. Although without the endogenous expression, HELF cells began to express EMMPRIN in a time-dependent manner after being co-cultured with SW480 cells.</p><p><b>CONCLUSIONS</b>Intercellular interactions between colon cancer cells and fibroblasts not only up-regulate the EMMPRIN expression in SW480 cells, but also induce its expression in HELF cells. Such interactions may play a crucial role in the invasion and metastasis of colon cancer.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of Chinese herbs for supplementing Shen and strengthening bone (HB) on myelogenic osteoclasts formation, and gene expression of interleukin-6 (IL-6), IL-6 receptor (IL-6R) and gp130 in bone marrow.</p><p><b>METHODS</b>Seventy-two healthy female SD rats of 3 months, were randomly divided into three groups, 24 in the sham-operated group (A), 24 in the ovariectomized group (B) and 24 in the after ovariectomy HB treated group (C). Bone marrow cells of 6 rats from each group were respectively collected and cultured at four time points (2nd, 4th, 6th and 12th weeks after operation). After 6 days of culture, the bone marrow cells were differentiated by Wright-Giemsa stain and TRAP stain, and total RNA in them was extracted by TRIZOL.</p><p><b>RESULTS</b>Beginning from the 2nd week, the osteoclasts formation in Group B was higher than that in Group A (P < 0.05), and IL-6, IL-6R gene expression significantly increased in Group B (P < 0.05 or P < 0.01). These changes reached the peak in the 4th to 6th week, with the level maintained to the 12th week. As for comparison of Group B and C, the above-mentioned changes were significantly weakened in the latter (P < 0.05 or P < 0.01). No significant change of gp130 gene expression revealed in the whole course in either group.</p><p><b>CONCLUSION</b>HB could inhibit the myelogenic osteoclasts formation in ovariectomized rats, this effect may be correlated with, partially at least, its inhibitory effect on the over-expressed IL-6 and IL-6R gene expression in myelocytes after ovariectomy.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study effects of insulin treatment on intracellular lipid content in livers and insulin resistance of type 2 diabetic rats.</p><p><b>METHODS</b>Type 2 diabetic rats were induced by injecting streptozotocin (25 mg/kg) and fat rich food. Then according to the results of the oral glucose tolerance test (OGTT) and glucose-induced insulin secretion test (IST), the rats were divided into two groups: control group (DC) and insulin treated group (DI). Normal rats (NC) served as controls. The treatment of each group with either NPH insulin (4 approximately 6 U . kg-1. d-1), or saline continued for 4 weeks. Body weight, OGTT, IST, blood lipids, intracellular lipids in liver and liver histology were studied. The insulin sensitivity index (ISI) was applied to assess the status of insulin resistance.</p><p><b>RESULTS</b>Blood lipid and intracellular lipids in livers in the DC were higher than those in NC (t = 2.59 approximately 15.77, P < 0.05) and the ISI was lower (t = 3.16, P < 0.05), with many fatty droplets appearing in the livers. In comparison to DC, DI showed that blood lipids were decreased, but lipids in livers were markedly increased (TG, TC, FFA increased 55.7%, 19.87%, 22.2%, respectively), and fatty droplets in hepatocytes were larger, but the ISI did not change significantly.</p><p><b>CONCLUSION</b>Insulin treatment can make blood glucose normal, increase the intracellular lipid content in the liver, and not increase the insulin resistance significantly.</p>
